We are thrilled to share our lab's involvement in a new study published in Nature Communications: "Epigenetic regulation of CD38/CD48 by KDM6A mediates NK cell response in multiple myeloma".
We tried to explain how epigenetic changes, particularly involving KDM6A, contribute to resistance against Daratumumab (Dara), an anti-CD38 monoclonal antibody widely used in multiple myeloma (MM) treatment. Loss of KDM6A reduces CD38 and CD48 expression via increased H3K27me3, leading to resistance to Daratumumab-mediated antibody-dependent cellular cytotoxicity (ADCC). Restoring sensitivity to Daratumumab is possible through EZH2 inhibitors, which upregulate CD38 and CD48.
These discoveries highlight new therapeutic possibilities for overcoming drug resistance in MM, including the potential integration of FDA-approved EZH2 inhibitors to enhance the efficacy of Daratumumab.
